Hypothesis: Symptomatic and asymptomatic knee osteoarthritis (OA) can be defined by clinical assessment, joint pathology and central pain processing.
Objectives
The overall aim of this study is to acquire post mortem tissue with matched demographic, clinical and pathologic data.
Study design
An open, exploratory study in which samples of excised post mortem tissues will be collected and harvested. The study population selected for this study comprises subjects whose bereaved spouse, relative or other next of kin has agreed to the donation of the deceased’s tissue at post mortem.
Sample and data acquisition/collection
Tissue obtained during the procedures below will be donated by the bereaved spouse, relative or other next of kin after full written informed consent has been obtained by a member of the research team:
• Knee joint tissues, muscle and local nerve fibres: obtained through anterior longitudinal incision.
• Human spinal cord and dorsal root ganglia: obtained via a posterior approach
Tissue will be collected at Sherwood Forest Hospitals NHS Foundation Trust. Samples may be processed and held at Sherwood Forest Hospitals NHS Foundation Trust and University of Nottingham, as frozen or fixed and embedded samples. Fresh tissue may be collected for in vitro studies.
All tissue will have matching datasets of demographic and clinical data.
Sample analysis
Samples will be used for a variety of research studies including biochemical, histochemical, and functional validation studies. Genetic tests may be carried out in order to determine whether genetic polymorphisms are associated with disease, and tissue may be used in research using laboratory animals. These tissues may also be used to form experimental cell lines which are a useful model for testing for example new therapies.
Start dates may differ between countries and research sites. The research team are responsible for keeping the information up-to-date.
