This is a study of osteoarthritis using tissue from patients knee joints removed at surgery for joint replacement. Patients with osteoarthritis will be asked to donate tissue and to provide a blood and urine sample for the study. Age matched patients without osteoarthritis, but who require limb amputation for other medical reasons, will also be asked to donate tissue, blood and urine for the study. This will provide important non-osteoarthritic samples for comparison in the study. Samples of tissue and cells from the OA and non-OA patients will be isolated for a range of analysis and live cells will also be grown in culture and tested. The main focus is on cartilage tissue and the chondrocytes, the live cells in cartilage. In addition cells from patients tissues, from urine and from blood, will be converted using gene transfer into stem cells (induced pluripotent stem cells) for further research into what makes cartilage become osteoarthritic. These studies require detailed comparison between OA and non-OA patients and combining the results from these different research approaches using the latest systems biology methods will provide a comprehensive analysis of osteoarthritis and it will identify many possible new routes to treatment.

OA Group: Patients for knee replacement with any nonosteoarthritis joint pathology, such as rheumatoid arthritis. Non-OA Controls: Patients requiring limb amputation, who have any joint pathology.
